In contrast with the Japanese oyster Crassostrea gigas and the Manila clam Ruditapes philippinarum, Ostrea edulis seed production in the hatchery has been reported to be erratic, with sudden and unexplained larval and post-metamorphosis mortalities. Fecundity and initial larval quality have been related to broodstock conditioning, but effects on larval development and metamorphosis remain poorly understood. In addition, molluscan larval mortalities have been often associated with bacterial contamination and flow-through techniques may help to overcome this problem. Both aspects have been considered in the present work. O. edulis broodstock were conditioned at 19 °C and fed three different microalgal diets. Two were single-species diets: Rhodomonas salina... |

Under controlled conditions of food density and temperature, larval performances (ingestion, growth, survival and settlement success) of the flat oyster, Ostrea edulis, were investigated using a flow-through rearing system. In the first experiment, oyster larvae were reared at five different phytoplankton densities (70, 500, 1500, 2500 and 3500 μm3 μL−1: ≈1, 8, 25, 42 and 58 cells μL−1 equivalent TCg), and in the second, larvae were grown at four different temperatures (15, 20, 25 and 30°C). Overall, larvae survived a wide range of food density and temperature, with high survival recorded at the end of the experiments. Microalgae concentration and temperature both impacted significantly larval development and settlement success. A mixed diet of Chaetoceros... |

Integration of shellfish farming along the French shoreline (around 5500 km) has been progressive, as marine bivalve culture is a traditional activity in our country. The history of mussel culture relates that the "bouchot" technique (intertidal culture on wooden poles, producing annually around 60,000 tons) was developed as early as the XIIIth century, by a refugee attempting to capture birds by sticking branches of brushwood along the shore. The European flat oyster, Ostrea edulis, already appreciated at the Roman period, was first cultivated in France after Victor Coste introduced the technique of spat collection at the end of the XIXth century. The Pacific cupped oyster, genus Crassostrea, now the most-cultivated in France (around 120,000 tons marketed... |

Bonamia ostreae is an intrahemocytic parasite that has been responsible for severe mortalities in the flat oyster Ostrea edulis since the 1970́s. The Pacific oyster Crassostrea gigas is considered to be resistant to the disease and appears to have mechanisms to avoid infection. Most studies carried out on the invertebrate immune system focus on the role of hemolymph, although mucus, which covers the body surface of molluscs, could also act as a barrier against pathogens. In this study, the in vitro effect of mucus from the oyster species Ostrea edulis and C. gigas on B. ostreae was investigated using flow cytometry. Results showed an increase in esterase activities and mortality rate of parasites exposed to mucus from both oyster species. In order to... |

The history of French oyster culture shows very well how fragile this farming is to stock overexploitation and diseases. The production of flat oyster, Ostrea edulis, fell dramatically after the appearance of two parasitic diseases: arteiliosis and bonamiosis. The curative means against bonamiosis (Bonamia ostreae) are restricted and imply a good knowledge of the parasite's life cycle and of its interaction with its host. Studying the expression of the parasite's and the host's genes during the infection seems essential in providing new adapted measures in order to maintain, if not relaunch the flat oyster culture. In this context, this thesis aims to understand the flat oyster-parasite interactions, and especially, the molecular basis of the resistance to... |

The first microscope observations of the digestive gland of Ostrea edulis were conducted by Comps (1970) and Herbach (1971), but the first effects had already been observed in the autumn of 1967. During summer 1968, an abnormal mortality occurred in the upstream part of the Aber Wrach, then spread downstream and into the Aber Benoît. Similar mortalities were also observed in the area of La Tremblade and in the Arcachon Bay. Since then, a permanent monitoring is conducted in the different Breton oyster culture centres and the Arcachon area. |

The European flat oyster Ostrea edulis has been considered as the traditional French oyster for a very long time and has now nearly disappeared from our farms, as a consequence of the two successive epizootic of the Marteilia refringens parasite and Bonamia ostreae. The actual quasi monoculture of Crassostrea gigas has made necessary the search for a diversification in oyster culture, including the search for a new start of the indigenous flat oyster culture. The Ifremer has been conducting parallel works since 1985 in order to better understand the biology of the parasite as well as to try and select different lines of flat oyster resistant to Bonamia ostreae. |

Survival, growth and fatty acid composition of Ostrea edulis larvae (L.) fed four different single species, microalgal diets, Tisochrysis lutea (T), Chaetoceros neogracile (Cg), Skeletonema marinoi (Sm) or Tetraselmis suecica (Ts) from broodstock to pre-settlement, were studied. Lower larval growth (5.5m to 6.5m/d) was recorded in progeny continuously fed single S.marinoi or T.suecica, whereas good growth was achieved with single T.lutea (7.8m/d). Larvae, originated from broodstock receiving Sm or Ts, exhibited growth compensation when fed a bispecific balanced diet (TCg). This did not occur when broodstock and larvae were fed Cg or T, for which single or mixed diets led to similar larval growth. Furthermore, survival was high (>90%) regardless of... |

Acid phosphatase activity was cytochemically detected at the ultrastructural level in the protozoan Bonamia ostreae, an intrahemocytic parasite of the flat oyster Ostrea edulis. Lead nitrate was used as capture agent for phosphate liberated during the hydrolysis of the subtrate (cytidine 5' monophosphate). Lead phosphate precipiation, indicative of enzyme activity, was located in membrane-bound organelles corresponding to the cytoplasmic inclusions known as "dense bodies". Acid phosphatase activity level and sodium L-tartrate inhibition were measured spectrophotometrically from a purified parasite suspension. The possible involvement of this enzyme in intracellular survival of B. ostrea is discussed. |
